Not necessarily. After Karl left, Luke streamed as Lara using the same clothes he was already wearing. If Karl has watched that after he left, then it’s not really hard for Karl to connect the dots that Luke and Lara are one and the same.
Still, that is a big “if”. That’s pretty likely, though, since he is already aware of Lara’s channel (even became a guest in one of the streams, actually). I do wish it was more explicitly shown that Karl knew about Luke being Lara, unless, of course, if that was done on purpose…

It is emplied that the secret Karl and Jackie know is the Lara/Luke secret as that is the only secret we know Luke has and they have never explicitly pointed out the secret these 2 know is anything but that. If it was just Luke feeling femme than they would have stated that more obvious I think.
The way they talk about it makes it seem like they only know Luke feeling femme, and not about Lara. Though, Luke did say that they both know his “deepest, darkest secret”, so I guess Luke just thought that Karl and Jackie would know about Lara, too.
That would actually make sense but unless that is established it’s headcannon.
